An endosymbiotic Pseudomonas sp. (MSI057), which could produce high yields of lipase, was isolated from marine sponge Dendrilla nigra, collected from the peninsular coast of India. Maximum production of enzyme was obtained in minimal medium supplemented with 1% tributyrin. Catabolite repression was observed when the medium was supplemented with readily available carbon sources. The optimum temperature and pH for the enzyme production was 30 degrees C and 9.0, respectively. The enzyme exhibited maximum activity in pH range of 8-9 with an optimum pH 9.0. The activity of purified enzyme was optimum at 37 degrees C and showed 80% activity at 20 degrees C and the enzyme activity decreased dramatically above 50 degrees C. Based on the present findings, the enzyme was characterized as psychrophilic alkaline lipase, which can be developed for industrial applications.  相似文献

Effect of biofouling on various polymers and composites such as, Polyurethane (PU), Silicone rubber (SR), Polyester (PET), Glass Fiber Reinforced Polymer (GFRP), Carbon fibre Reinforced Plastic (CFRP) and Syntactic foams (SF) deployed for a period of one year in marine waters at a depth of 1 m was studied. These materials find wide marine applications. SR with lowest surface energy was the least fouled. Maximum barnacle attachment was seen on hard surface (GFRP) and minimum on flexible surface (SR). Attachment of barnacles and polychaetes are positively correlated with surface energy. Fouling load is positively correlated with Surface energy and hardness. The surface energy, hardness and tensile strength reduced while surface roughness considerably increased during this period. Maximum gravimetric weight loss was seen in PET (7.49%) followed by PU (4.25%) and minimum in CFRP (0.45%). Maximum thermogravimetric weight loss was observed in PET (73.5% at 400 °C) followed by PU (71.1%) and least in SR (2.4%). Fourier Transform infrared spectrum revealed that carbonyl/oxidation indices decreased for PET, GFRP, CFRP, and SR indicating biotic degradation. The same index increased for PU indicating abiotic oxidation.  相似文献

Few reliable data exist concerning the recurrence risk for individual trisomies or the risk for recurrence of trisomy for a different chromosome. We collected records from two sources: (1) prenatal diagnoses performed at the Hopital Sainte-Justine in Montreal and (2) karyotype analyses performed at Genzyme. Using the standardized morbidity ratio (SMR), we compared the observed number of trisomies at prenatal diagnosis with the expected numbers, given maternal age-specific rates (by single year). SMRs were calculated both for recurrence of the same trisomy (homotrisomy) and of a different trisomy (heterotrisomy). After all cases with an index trisomy 21 were combined, the SMR for homotrisomy was 2.4 (90% CI 1.6-3.4; P=.0005). For women with both the index trisomy and subsequent prenatal diagnosis at age <30 years, the SMR was 8.0; it was 2.1 for women with both pregnancies at age >/=30 years. For the other index viable trisomies (13, 18, XXX, and XXY) combined, the SMR for homotrisomy was 2.5 (90% CI 0.7-8.0). For heterotrisomy, the SMR after an index trisomy 21 was 2.3 (90% CI 1.5-3.8, P=.0007); the SMR did not vary with maternal age at the first trisomy. When all cases with index viable trisomies were combined, the SMR for heterotrisomy was 1.6 (90% CI 1.1-2.4; P=.04). For prenatal diagnoses following a nonviable trisomy diagnosed in a spontaneous abortion (from Genzyme data only), the SMR for a viable trisomy was 1.8 (90% CI 1.1-3.0; P=.04). The significantly increased risk for heterotrisomy supports the hypothesis that some women have a risk for nondisjunction higher than do others of the same age.  相似文献

Conformational preferences of 1,4,7-trithiacyclononane were studied using a highly efficient sampling technique based on local nonstochastic deformations and the MM2(91) force field. The results show that conformers that the molecule adopts in the crystal state were found to be low-energy conformers (LECs) within 5 kcal mol(-1) of the global minimum. A conformation with C1 symmetry was the global minimum and the C3 and C2 conformations were calculated to be 0.03 and 1.78 kcal mol(-1) higher in energy, respectively. The structures were further minimized using Density Functional Theory (DFT) calculations with two different functionals. The C2 and the C1 conformations were found to be LECs with the C3 conformation more than 4.0 kcal mol(-1) above the global minimum. The relative energies and structural ordering obtained using the BP86 functional are in agreement with the previously reported relative energies calculated using second-order Moller-Plesset (MP2) ab initio calculations. With the energy ordering being dependent on the molecular mechanics force field used, the approach of MM-->DFT (searching exhaustively the available conformational space at the MM level followed by generating the energy ordering through DFT calculations) appears to be appropriate for thiacrown ethers.  相似文献

The SED1 gene (YDR077W), coding for the major cell wall glycoprotein of Saccharomyces cerevisiae stationary-phase cells, contains two blocks of tandem repeat units located within two distinct regions of the nucleotide sequence. A PCR survey of the SED1 open reading frames (ORFs) of 186 previously uncharacterized grape must isolates of S. cerevisiae yielded 13 PCR profiles arising from different combinations of seven SED1 length variants in individuals homozygous or heterozygous for the gene. Comparison of the nucleotide sequences of a group of representatives of each of the seven length variants with those of S288C and the type strain, CBS1171, unequivocally identified them as SED1 alleles and provided evidence for the presence of two minisatellite-like sequences, variable in length, within the ORF of an S. cerevisiae gene. The segregation analyses of the SED1 length variants and other genetic markers in 13 isolates representative of each PCR profile suggested that molecular mechanisms involved in minisatellite expansion and contraction may be responsible for SED1 heterozygosities within a population of homothallic must isolates of S. cerevisiae.  相似文献

Summary Thermophilic bacterial leaching of low-grade manganese oxide ore was, demonstrated at 50°C and 70°C. A static culture system was used with cane molasses as the nutrient source. Nitrogen fertilization of roots enhances shoot growth in plants and cytokinins are known to initiate bud outgrowth in shoots. Is it possible that root-derived cytokinins may play a role in long-distance signaling for nitrogen availability in the promotion of sylleptic branching in hybrid poplar? Nitrogen fertilization in the form of 5 mM NH4NO3, KNO3 or NH4Cl was applied to roots of three hybrid poplar clones exhibiting contrasting degrees of sylleptic branching. Cytokinin (0.1-1 mM benzyladenine, BA) was applied directly to lateral buds of shoots. Glutamate, asparagine and glutamine were also applied as drops to buds or as foliar sprays. NH4NO3, KNO3 and NH4Cl all usually enhanced sylleptic branching within a week in the high sylleptic clone (11-11) but in four out of five trials there was no effect in the low sylleptic clone (47-174). NH4NO3 added directly to buds had no effect. Also, glutamate, asparagine and glutamine had no effect. However, 1 mM BA promoted lateral bud outgrowth in all three clones. These results are consistent with the long-distance nitrogen signaling hypothesis of Forde and Sakakibara wherein nitrogen is transduced to cytokinin via enhanced ipt activity in the roots and is translocated up the shoot with the subsequent promotion of leaf/bud outgrowth.  相似文献

We analyzed mycorrhizal types and dark septate endophyte (DSE) fungal associations in a shola vegetation of Western Ghats region, southern India. Plants belonging to 29 species of 19 families were assessed for mycorrhizal type and DSE fungal association. Five mycorrhizal classes were classified based on morphological traits: arbuscular, ecto-, ectendo-, ericoid-, and orchid mycorrhizas. Arbuscular mycorrhizal (AM) association was the most predominant mycorrhizal type, occurring in 16 plant species, followed by orchid (3 species), ericoid- (2 species), and ecto- and ectendomycorrhizas (1 species each). Mycorrhizal association is reported for the first time in 19 plant species. DSE fungal association was found in six plant species. Arum- and Paris-type AM morphology was found, respectively, in 10 and 5 plant species, with intermediate type recorded in one species. In this study, some new records on the morphological types of AM in some plant families were obtained. Further occurrence of ectendomycorrhizas in Pinus oocarpa and dark septate fungal association in Eleaocarpus munronii, Symplocos cochinchiensis, Daphniphyllum neilgherrense, Euodia roxburghiana, Syzygium arnottianum, and Syzygium montanum are reported for the first time. Roots of Berberis tinctoria, Mahonia leschenaultii (Berberidaceae), Elaeagnus latifolia (Elaeagnaceae), and Elaeocarpus oblongus (Elaeocarpaceae) lacked any fungal structures.  相似文献
